This study investigated two outbreaks of spontaneous poisoning by Baccharis coridifolia (Asteraceae) in early-weaned beef calves in Tacuarembó, Uruguay. A total of 34 affected calves showed signs of salivation, anorexia, apathy, marked dehydration, and diarrhea. Deaths occurred 36-72 h after consumption and mortality varied from 37.5% to 43.3% for outbreak 1 and outbreak 2, respectively. The main pathological findings include diffuse severe necrosis of the prestomachs and lymphoid tissues. Ultrastructurally, epithelial cells of the rumen showed swelling, lysis of the organelles, degradation of intercellular attachments, and degradation of the nuclear chromatin. Using LC-MS with diagnostic fragmentation filtering, 56 macrocyclic trichothecenes including glycosyl and malonyl conjugates were identified. The total concentration of macrocyclic trichothecenes, including conjugates, was estimated to be 1.2 ± 0.1 mg/g plant material. This is the first report of these malonyl-glucose conjugates from Baccharis coridifolia.

BACKGROUND: In humans, muscle-invasive bladder cancer (MIBC) is highly aggressive and associated with a poor prognosis. With a high mutation load and large number of altered genes, strategies to delineate key driver events are necessary. Dogs and cats develop urothelial carcinoma (UC) with histological and clinical similarities to human MIBC. Cattle that graze on bracken fern also develop UC, associated with exposure to the carcinogen ptaquiloside. These species may represent relevant animal models of spontaneous and carcinogen-induced UC that can provide insight into human MIBC. RESULTS: Whole-exome sequencing of domestic canine (n = 87) and feline (n = 23) UC, and comparative analysis with human MIBC reveals a lower mutation rate in animal cases and the absence of APOBEC mutational signatures. A convergence of driver genes (ARID1A, KDM6A, TP53, FAT1, and NRAS) is discovered, along with common focally amplified and deleted genes involved in regulation of the cell cycle and chromatin remodelling. We identify mismatch repair deficiency in a subset of canine and feline UCs with biallelic inactivation of MSH2. Bovine UC (n = 8) is distinctly different; we identify novel mutational signatures which are recapitulated in vitro in human urinary bladder UC cells treated with bracken fern extracts or purified ptaquiloside. CONCLUSION: Canine and feline urinary bladder UC represent relevant models of MIBC in humans, and cross-species analysis can identify evolutionarily conserved driver genes. We characterize mutational signatures in bovine UC associated with bracken fern and ptaquiloside exposure, a human-linked cancer exposure. Our work demonstrates the relevance of cross-species comparative analysis in understanding both human and animal UC.

This report describes 2 events of degenerative myelopathy in 4- to 27-day-old piglets, with mortality rates reaching 40%. Sows were fed rations containing low levels of pantothenic acid. Piglets presented with severe depression, weakness, ataxia, and paresis, which were more pronounced in the pelvic limbs. No significant gross lesions were observed. Histologically, there were degeneration and necrosis of neurons in the spinal cord, primarily in the thoracic nucleus in the thoracic and lumbar segments, and motor neurons in nucleus IX of the ventral horn in the cervical and lumbar intumescence. Minimal-to-moderate axonal and myelin degeneration was observed in the dorsal funiculus of the spinal cord and in the dorsal and ventral nerve roots. Immunohistochemistry demonstrated depletion of acetylcholine neurotransmitters in motor neurons and accumulation of neurofilaments in the perikaryon of neurons in the thoracic nucleus and motor neurons. Ultrastructurally, the thoracic nucleus neurons and motor neurons showed dissolution of Nissl granulation. The topographical distribution of the lesions indicates damage to the second-order neurons of the spinocerebellar tract, first-order axon cuneocerebellar tract, and dorsal column-medial lemniscus pathway as the cause of the conscious and unconscious proprioceptive deficit, and damage to the alpha motor neuron as the cause of the motor deficit. Clinical signs reversed and no new cases occurred after pantothenic acid levels were corrected in the ration, and piglets received parenteral administration of pantothenic acid. This study highlights the important and practical use of detailed neuropathological analysis to refine differential diagnosis.

The existing rearing protocols for Anastrepha fraterculus must be reviewed to make economically viable the production of sterile flies for their area-wide application. Additionally, evidence of sexual incompatibility between a long-term mass-reared Brazilian strain and wild populations has been found. To address these issues, this study aimed to refine rearing protocols and to assess the suitability of an A. fraterculus strain for the mass production of sterile flies. A series of bioassays were carried out to evaluate incubation times for eggs in a bubbling bath and to assess the temporal variation of egg production from ovipositing cages at different adult densities. A novel larval diet containing carrageenan was also evaluated. Egg incubation times higher than 48 h in water at 25 °C showed reduced larval and pupal yields. Based on egg production and hatchability, the density of 0.3 flies/cm2 can be recommended for adult cages. The diet with carrageenan was suitable for mass production at egg-seeding densities between 1.0 and 1.5 mL of eggs/kg of diet, providing higher insect yields than a corn-based diet from Embrapa. Even after two years of being reared under the new rearing protocols, no sexual isolation was found between the bisexual strain and wild flies.

Dorsal-ventral patterning of the Drosophila embryo depends on the NFκB superfamily transcription factor Dorsal (Dl). Toll receptor activation signals for degradation of the IκB inhibitor Cactus (Cact), leading to a ventral-to-dorsal nuclear Dl gradient. Cact is critical for Dl nuclear import, as it binds to and prevents Dl from entering the nuclei. Quantitative analysis of cact mutants revealed an additional Cact function to promote Dl nuclear translocation in ventral regions of the embryo. To investigate this dual Cact role, we developed a predictive model based on a reaction-diffusion regulatory network. This network distinguishes non-uniform Toll-dependent Dl nuclear import and Cact degradation, from the Toll-independent processes of Cact degradation and reversible nuclear-cytoplasmic Dl flow. In addition, it incorporates translational control of Cact levels by Dl. Our model successfully reproduces wild-type data and emulates the Dl nuclear gradient in mutant dl and cact allelic combinations. Our results indicate that the dual role of Cact depends on the dynamics of Dl-Cact trimers along the dorsal-ventral axis: In the absence of Toll activation, free Dl-Cact trimers retain Dl in the cytoplasm, limiting the flow of Dl into the nucleus; in ventral-lateral regions, Dl-Cact trimers are recruited by Toll activation into predominant signaling complexes and promote Dl nuclear translocation. Simulations suggest that the balance between Toll-dependent and Toll-independent processes are key to this dynamics and reproduce the full assortment of Cact effects. Considering the high evolutionary conservation of these pathways, our analysis should contribute to understanding NFκB/c-Rel activation in other contexts such as in the vertebrate immune system and disease.

RESUMO

In a litter of three puppies, one was stillborn and had facial and brain defects. Fusion of the maxilla and mandible and absence of the face were observed. The forebrain (telencephalon and the diencephalon) was reduced in size and fused, and the telencephalic longitudinal fissure, olfactory bulbs, and optic nerves were absent (Figures 6 and 7). Lissencephaly was observed in the telencephalon and cerebellum. A diagnosis of aprosopia/holoprosencephaly was made.

ABSTRACT: Acute hepatotoxicity caused by plants poisoning is responsible for economic losses in farm animals in Brazil. Reports of Cestrum intermedium natural poisoning in cattle are not commonly described in Rio Grande do Sul (RS). This study aimed to document an outbreak of spontaneous C. intermedium poisoning in dairy cattle in the Central-Eastern Mesoregion of RS. Three nine-month-old Holstein and Jersey heifers were affected after they were placed in a small paddock with shortage forage. In this area, specimens of C. intermedium Sendtn with signs of consumption were observed. Morbidity and lethality rates were 100% and clinical courses ranged from 9 to 12 hours. At post mortem examination of the three heifers, there was predominance of acute liver lesions. The liver was moderately enlarged and on the cut surface there was a marked accentuation of the lobular pattern and hemorrhage. Inside the rumen, partially digested C. intermedium Sendtn leaves were observed. The histological aspects of the liver were mostly centrilobular coagulative necrosis and hemorrhage, frequently extended to the midzonal region. The immunohistochemistry technique was performed, in which the polyclonal antibody caspase 3 was used in liver fragments. Moderate to marked immunolabeling was observed in the cytoplasm and nucleus of hepatocytes, predominantly on the periphery of areas of hepatic necrosis indicating cell apoptosis. The diagnosis of C. intermedium Sendtn poisoning in dairy cattle in this study was based on epidemiological, clinical and anatomopathological findings. Since the C. intermedium poisoning is uncommon in dairy cattle, we are describing it for the first time in the Central-Eastern Mesoregion of RS, and represents a differential diagnosis of other acute toxic liver diseases in cattle.

Relatos de intoxicações naturais por Cestrum intermedium em bovinos não são comumente descritos no Rio Grande do Sul (RS). Este trabalho teve como objetivo documentar um surto de intoxicação espontânea por C. intermedium em bovinos leiteiros ocorrido na região Centro-Oriental do RS. Três bovinos com nove meses de idade, das raças Holandês e Jersey, foram acometidos depois de serem introduzidos em um pequeno piquete com escassez de forragem. Nessa área foram observados exemplares de Cestrum intermedium Sendtn com sinais de consumo. As taxas de morbidade e letalidade foram de 100% e os cursos clínicos variaram de 9 a 12 horas. Na necropsia dos três bovinos, observou-se predomínio de lesões hepáticas agudas. O fígado dos animais acometidos estava moderadamente aumentado de tamanho e na superfície de corte havia marcada acentuação do padrão lobular e hemorragia. Na abertura do rúmen, foram observadas folhas de Cestrum intermedium Sendtn parcialmente digeridas. Histologicamente, o fígado apresentava necrose coagulativa centrolobular e hemorragia. Frequentemente, essas alterações se estendiam para a região mediozonal. Foi realizada a técnica de imuno-histoquímica, na qual utilizou-se o anticorpo policlonal caspase 3 em fragmentos de fígado. Notou-se marcação moderada a acentuada em citoplasma e núcleo de hepatócitos, predominantemente na periferia das áreas de necrose hepática indicando apoptose celular. O diagnóstico de intoxicação por Cestrum intermedium Sendtn nos bovinos leiteiros desse estudo, foi realizado com base nos achados epidemiológicos, clínicos e anatomopatológicos. A intoxicação por C. intermedium é incomum em bovinos leiteiros, sendo descrita pela primeira vez na região centro oriental do RS e representa um diagnóstico diferencial de outras hepatopatias tóxicas agudas.

ABSTRACT: The shape is one of the key features of a lesion and a pathologist must be able to identify and interpret these forms in the context of any gross and microscopic changes. One of the principles of adult learning is to engage the learner with previously understood information. If, when presenting material that is new, a connection with something the student already has familiarity with, the learning process is accelerated. As the learners are already familiar with shapes they have encountered throughout their pre-pathology learning, these can be used to hasten the incorporation and understanding of lesions. This paper describes various shapes that are used in describing lesions in veterinary pathology.


RESUMO: A forma é uma das principais características de uma lesão. Um patologista deve ser capaz de identificar e interpretar essas formas no contexto de quaisquer alterações macroscópicas ou microscópicas. Um dos princípios da aprendizagem de adultos é envolver o aluno com informações previamente compreendidas. Se, ao apresentar um material novo, se estabelece uma conexão com algo que o aluno já conhece, o processo de aprendizagem torna-se mais rápido e eficiente. Como os alunos já estão familiarizados com as formas que encontraram ao longo de sua aprendizagem pré-patologia, elas podem ser usadas para acelerar a incorporação e a compreensão das lesões. Este artigo descreve várias formas que são usadas na descrição de lesões em patologia veterinária.

ABSTRACT: Histopathology is an old science that is still currently utilized for disease diagnosis and research. The routinely processed histologic slides stained with hematoxylin and eosin are still used worldwide in most if not every histopathology laboratory. The technique is inexpensive, quick to perform, and allows the diagnosis of a fantastic variety of tissue changes and diseases. Skills in description and interpretation in histopathology are a craft that can be learned by repeatedly and systematically observing simple rules. In this article, we offer a few advices to help trainees in veterinary pathology at the start of their careers. Those advices are drawn from our experience in the diagnostic pathology routine and from the veterinary pathology literature. To enhance the understanding of these important steps in the histopathologic description of tissues, we decided to illustrate most concepts expressed here. We hope that our effort can add a bit to the development of future pathologists. Just like Alice, let us follow the White Rabbit into his burrow for this challenging experience!

A técnica não é cara, é de execução rápida e permite o diagnóstico de uma fantástica variedade de doenças. As habilidades em descrever e interpretar os achados histopatológicos é um ofício que pode ser aprendido pela observação repetida e sistemática de regras simples. Para ajudar os estudantes de patologia no início de suas carreiras, abordamos aqui algumas dessas regras, extraídas tanto de nossa experiência quanto da literatura relacionada à patologia veterinária. Para aumentar a compreensão dos tópicos, decidimos ilustrar praticamente todos os conceitos expressados neste manuscrito. Esperamos que nosso esforço possa contribuir um pouco para o desenvolvimento de aspirantes a patologistas. We describe a case of penile tuberculosis in a bull. The prepuce was thickened, firm and pale with multifocal to coalescent caseous nodules, and the inguinal and mesenteric lymph nodes were moderately enlarged. Lesions in the prepuce, penis and lymph nodes were characterized by multifocal to coalescent areas of caseous necrosis and marked granulomatous inflammation. Acid-fast bacilli were seen within necrotic foci and Langhans giant cells. Mycobacteriumtuberculosis var. bovis was identified in inguinal lymph nodes by the polymerase chain reaction technique and by bacterial isolation. Although rare, tuberculosis should be considered in the differential diagnosis of granulomatous lesions in the genital tract of bulls.

The clinical and pathological findings of a case of fatal disseminated toxoplasmosis in a captive brown-throated sloth (Bradypus variegatus) from the northern region of Brazil are reported. Clinical signs were nonspecific and included apathy, prostration, dyspnoea, and loss of appetite. Treatment with penicillin was attempted, but the animal died within five days of the onset of clinical signs. Microscopically, there was acute inflammation in the liver, spleen, and lungs associated with necrosis and a few cysts and extracytoplasmic tachyzoites, with a morphology compatible with Toxoplasma gondii. Tissue sections were submitted for immunohistochemistry that confirmed T. gondii as the aetiological agent. To the authors' knowledge, this is the first report of toxoplasmosis in B. variegatus.

RESUMO

Adult learning, or andragogy, provides a novel way of appreciating using food analogies as an effective learning tool in veterinary pathology. Facilitation of adult learning requires that new concepts be presented in a way that draws on the learner's experience. Because veterinary students will have had considerable experience with a plethora of food items prior to enrolling in a pathology course, food analogies can provide an easy conduit for incorporating key learning concepts regarding veterinary pathology. In this paper, many of these analogies are presented, along with the mechanisms responsible for each of the characteristic lesions, in the hopes that their usefulness in the classroom can be highlighted to create a more engaging and facilitated learning environment.(AU)


A aprendizagem de adultos, ou andragogia, é uma nova maneira de apreciar o uso de analogias de alimentos como uma ferramenta eficaz no aprendizado em patologia veterinária. A facilitação da aprendizagem de adultos requer que novos conceitos sejam apresentados de uma forma que se baseie na experiência do aluno. Como os estudantes de medicina veterinária já terão sido expostos a um número considerável de tipos de alimentos antes de se matricularem na disciplina de patologia, as analogias de alimentos podem fornecer um ótimo conduto para incorporar os conceitos-chave na aprendizagem da patologia veterinária. Neste artigo, muitos dessas analogias são apresentadas juntamente com os mecanismos responsáveis por cada uma das lesões características, na esperança de que sua utilidade na sala de aula possa ser destacada para criar um ambiente de aprendizado mais envolvente e favorável.(AU)

Toxoplasmosis is a disease with a worldwide distribution that affects a wide variety of animal species, though with rare descriptions in chickens. We describe the clinical, epidemiological, pathological, and molecular aspects of a toxoplasmosis outbreak in domestic chickens and guinea fowl in southern Brazil. The flock was composed of 47 domestic chickens and 29 guinea fowl. Of these, 22 birds showed clinical signs of lethargy, anorexia, and neurological signs over a clinical course of 24-72 h, and 15 died. Epidemiological data were obtained through fieldwork performed at the chicken farm and necropsies of six birds. Gross lesions were absent at necropsy, and histopathological findings included inflammatory infiltrate of macrophages, lymphocytes, and plasma cells and necrosis in several tissues associated with intralesional Toxoplasma gondii. Immunohistochemistry for T. gondii was positive. Additionally, restriction fragment length polymorphism (RFLP) analysis with 11 markers (SAG1, SAG2 (5'3'SAG2 and alt. SAG2), SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, Apico, and CS3) and microsatellite (MS) analysis with 15 markers (TUB2, W35, TgMA, B18, B17, M33, IV.1, XI.1, N60, N82, AA, N61, N83, M48, and M102) were performed. PCR-RFLP revealed T. gondii genotype ToxoDB-PCR-RFLP #280, and MS analysis also showed a unique genotype. This is the first description of this genotype in chickens and adds to the evidence suggesting considerable genotypic diversity of T. gondii in Brazil.

Dr. Jürgen Döbereiner was born in Germany, on the 1st of November 1923, and lived in Brazil for 68 years during which time he developed a range of scientific projects in veterinary pathology and related disciplines. His main interests were the identification of new poisonous plants and mineral deficiencies and the causes of "cara inchada" ("swollen face" a periodontal disease) and botulism in livestock. This research has resulted in the improved health and saving of hundreds of thousands of animals, mainly cattle, annually, and is consequently of enormous economic value to the country. This contribution remains largely under appreciated. He was also involved in organizing diagnostic methods for identifying infectious diseases such as African swine fever and glanders in horses. One of his other major achievements has been the foundation and editing of specialized scientific journals for the documentation of veterinary science research results. At the beginning of his career in the 1950s, he and colleagues from the Institute for Animal Biology (IBA) were struggling to find a national scientific journal where research results from veterinary medicine could be published with practical application to the Brazilian reality. In consequence, the team founded "Arquivos do Instituto de Biologia Animal" and published three volumes (1959-1961). He then founded and edited "Pesquisa Agropecuária Brasileira" (The Brazilian Journal of Agricultural Research") that included a veterinary section. A series of veterinary volumes were published (1966-1976). Finally, in 1978 he helped create the Brazilian College of Veterinary Pathology (CBPA) that published "Pesquisa Veterinária Brasileira" (The Brazilian Journal of Veterinary Research) from 1981. The main goal was to communicate the most relevant disease problems of Brazilian livestock, in particular pathology and related subjects such as epidemiology, clinical study series and laboratory diagnosis to field veterinarians and academics. Dr. Jürgen Döbereiner was president of CBPA (1978-2018) and chief editor of "Pesquisa Veterinária Brasileira" (1981-2018). He passed away on the 16th of October, 2018, at the age of 94 at his home in Seropédica/RJ, Brazil.(AU)


Dr. Jürgen Döbereiner nasceu na Alemanha em 1 de novembro de 1923, durante 68 anos viveu no Brasil e desenvolveu trabalhos científicos no campo da patologia veterinária latu sensu. Sua contribuição científica de destaque foi em temas como plantas tóxicas de interesse pecuário, deficiências minerais em animais de produção, cara inchada (doença periodontal) dos ruminantes, botulismo e diagnóstico de doenças infecciosas. Estas pesquisas resultaram na melhoria da saúde e de centenas de milhares de animais, principalmente bovinos e, consequentemente, foram de enorme valor econômico para o país. Esta contribuição ainda permanece em grande parte subestimada. De grande destaque para a ciência brasileira foi ainda a sua atuação profissional na documentação científica de resultados de pesquisa. No início de sua carreira na década de 1950, Dr. Döbereiner e outros pesquisadores do Instituto de Biologia Animal (IBA) detectaram a necessidade de um periódico científico nacional para publicar resultados de pesquisas com aplicação pratica à realidade brasileira. Dessa iniciativa surgiram os Arquivos do Instituto de Biologia Animal, que publicou três fascículos (1959-1961), em seguida o Dr. Jürgen Döbereiner participou na fundação da revista Pesquisa Agropecuária Brasileira que publicou a Série Veterinária (1966-1976) e finalmente em 1978, houve a fundação do Colégio Brasileiro de Patologia Animal (CBPA) que publica desde 1981 a revista Pesquisa Veterinária Brasileira. Este periódico científico foi criado para apresentar à comunidade, principalmente veterinários de campo e professores, os principais problemas de saúde em animais de produção no Brasil, ou seja, patologia em seu sentido amplo, envolvendo as áreas de epidemiologia, clínica e diagnóstico laboratorial. Dr. Jürgen Döbereiner, que foi presidente do CBPA (1978-2018) e Editor-Chefe da revista Pesquisa Veterinária Brasileira (1981-2018), faleceu em casa, em 16 de outubro de 2018, aos 94 anos, no município de Seropédica/RJ.(AU)

Toxic plants containing monofluoroacetate (MFA) cause sudden death in livestock in Australia, South Africa and Brazil, causing economic losses to producers. The objective of this study was to determine the amount of MFA present in young leaves, mature leaves, senescent leaves, and seeds of Amorimia pubiflora harvested at different times of the year and to determine their toxic effect on sheep. Samples of Amorimia pubiflora were collected during April, August and December of 2015 and March of 2016, separated according to the vegetative stage (young leaves, seeds, mature leaves, and senescent leaves), dried in an oven, and administered in daily doses of 5 g/kg/body weight (bw) of fresh leaves to sheep through ruminal cannulae. The experiment was divided into four stages according to the time of collection of the plant so that each sheep received a different vegetative stage of the plant (young leaves, mature leaves, and senescent leaves). Only in the second stage of the experiment was it possible to collect A. pubiflora seeds, which were administered using the same method used for the administration of the leaves. The sheep were dosed with the plant until they showed clinical signs of toxicosis or until the plant was no longer available. Aliquots of leaves and seeds of A. pubiflora were analyzed for MFA concentration. The seeds and young leaves had higher concentrations of MFA than did the mature (harvested in August and December) and senescent (harvested in December) leaves. However, all vegetative stages of the plant were toxic and caused fatal poisoning. The results of our study showed that A. pubiflora is toxic to sheep even when MFA concentrations are low, demonstrating that the presence of this substance is a risk factor for the occurrence of poisoning. Knowing the toxic principle and its variations allow us to determine the conditions for the occurrence of plant toxicosis as well as possible treatment, control, and prophylaxis methods, contributing significantly to the reduction of economic losses on farms due to plant poisoning.
